Vacuum compartment backpack
I have a backpack that has a vacuum compartment where I can put clothes and then vacuum pack them with a hand held compressor. This allows me to get a large number of clothes in my pack.
my question is, how likely with I be asked to open the seal during customs??? It will be a mess if I have to.

Welcome to Fodors. Were are you flying? Domestic or International? I suspect international because you mention Customs
But never the less . . . IF you have nothing suspicious in your bag the chances are slim that Customs or TSA or other agencies will ask to open it. Make sure there is only soft goods/clothing in that compartment and it shouldn't be an issue. But nothing is ever guaranteed -- they can always ask.
I occasionally use a Space Bag™ (same concept -- vacuum bag) in my carry on and it has never once been questioned. Though I don't pack anything weird or suspicious looking.

It really depends where you are going, some places are really rigorous. Mexico is, which is crazy as they have so many drugs themself, but that was the only airport where I've had to have my bag examined for no reason whatsoever. So if you are going to Mexico, it could happen. If you have anything smelly at all, it could also happen as a dog may sniff it out. I've seen that happen (they were carrying something illegal, by the way). But do not carry any plant items, etc.

If the contents of the bag get too dense for TSA to see inside then they may want you to open it.... Doubtful, but you never know. I would not put any dense /non compressible items into that compartment. One trip they didn't like a block of cheese I was carrying that looked like it was connected to some wires (headphones).... Go figure.
Customs outside of the US hasn't looked into my bags since travel to the USSR in the 60's.
Customs entering the US has looked in my bags from time to time but not pulled things apart.
